Illumina Genome Analyzer

Lab Introduction and Consultation

The Genomics Resource has recently acquired a state-of-the-art, high-throughput sequencing-by-synthesis (HT-SBS) Illumina Genome Analyzer System, consisting of an Illumina Cluster Station and an Illumina Genome Analyzer. This technology was originally developed by Solexa, Inc. and is often referred to as 'Solexa Sequencing.' The Cluster Station generates up to 40 million template clusters onto a single flow-cell surface. Each flow-cell contains 8 individual lanes, of which one lane is dedicated to a control sample. The flow-cell is introduced into the Genome Analyzer, which performs massive parallelization of up to 35 nucleotide reads. This process enables the sequencing of over 1 billion bases per 72 hour run. We will be upgrading the Illumina Genome Analyzer in late Spring 2008 so that we can perform paired-end sequencing, as well.

The Illumina sequencing system can be used for a variety of purposes including DNA re-sequencing, small RNA analysis, ChIP-seq and digital expression analysis. An excellent overview of Illumina's Genome Analyzer System and applications is available at their website.

The following are instructions on how Cancer Consortium members can access Illumina Genome Analyzer sequencing services at FHCRC. Those new to this technology or new to using the Genomics Resource are strongly encouraged to meet with lab personnel prior to planning a project. For non-consortium members interested in services please contact the Genomics Resource at (206) 667-2714.

Reagents and Consumable Purchases

Each investigator is required to purchase the appropriate kits for sample preparation, cluster generation, and sequencing directly from Illumina. Current pricing is available. Please note that reagent shelf life is 6 months and accordingly you should not make purchases until you've reserved time for your project with lab personnel (Andy Marty at jamarty@fhcrc.org or (206) 667-1393)
