Operations of Electron Microscopes

Training is provided on the electron microscope for those investigators who prefer to do their own scoping. The hands-on training includes loading samples, instruction on critical focus, light intensity and image recording.Training is provided by appointment only, at no cost.

Basic TEM:
This class will detail preparation and processing of biological specimens for TEM. Included in this class are fixation, dehydration, and embedding processes. Specimen support films, sectioning, and negative staining are also addressed. This class will also give an overview of instrumentation (TEM microscopes) and support equipment. A procedures manual with specific protocols and resources will be provided with this class.

Basic SEM:
This class will detail preparation and processing of biological specimens for SEM. Included in the class are fixation, dehydration, and drying methods (HMDS or CPD). Mounting and sputter coating will also be addressed. This class will also give an overview of instrumentation (SEM microscope) and support equipment. A procedures manual with specific protocols and resources will be provided with this class.

Cryo Techniques:
This class will explain how to prepare samples for high pressure freezing (HPF) and demonstrate the process. The class will address automatic freeze substitution and cryo-ultramicrotomy. An overview of immunoelectron microscopy will also be included.
